0% equals sacrificing at same amount.
50% equals 1.5 times your current amount.
-50% equals half your current amount.

what effect will bring the change from 1/2 to e.g. 1/5 or 5/10 ?

It is a random between those 2.
So 1/5 would mean the arena manager will only be played every 1-5 Minutes. So e.g.
1. 11:53
2. 11:58
3. 12:01
4. 12:02

etc.

You could set it e.g. to like 10-90 which would result in "more" human behaviour as then it would have certain "breaks".

Or as the other stuff is running anyways you can even set it to 0 to do arenamanager in each cycle (last colum in Account List shows current cyclic time)

Is there still a interference between Arena-Manager and Fortress fights? Like if the bot building stuff in the Arena-Manager like with lowest delays 0/0 will it have still consequences for fortress?

As long as you keep a limititation for the maximum build commands, you won´t enter a dead loop where the Arena Manager keeps building for ages, before it let´s the bot perform the rest.
You can easily check this by looking into the cyclic time. Normal is a value between 12 and 50s. So in case of it beeing higher consider lowering the maximum allowed build comamnds per cycle.

What settings can we use to ensure the bot will build immediately after sacrifice?
Right now it takes 20 minutes to it to build after a sacrifice.

For me it's starting building asap after sacrificed
